Thecal and interstitial cells in polycystic ovaries (PCO) in the rat

Abstract: Two different types of experimentally-induced polycystic ovaries (PCO) have been examined. A macrocystic ovarian condition is induced by estradiol valerate (EV) injection, whereas a microcystic ovarian condition is engendered with subcutaneous estradiol implants. In both of these models thecal and secondary interstitial cells were characterized using three functionally significant indices. Expression of alkaline phosphatase was evaluated immunohistochemically, hCG/LH-binding capacity was assessed by means of E… Show more

“…Afterwards, outgrowing granulosa cells also start to mingle in between, leading to a mixed monolayer of both somatic cell types. To identify theca cells and discriminate them from granulosa cells, alkaline phosphatase (AP) activity was detected [speciWc for theca interna cells (Convery and Brawer 1991)], using a Xuorogenic substrate. After removal of conditioned medium, cultured follicles were Wxed with ice-cold acetone (¡20°C) within the culture wells and incubated for 45 min in a mixture of naphthol AS-BI phosphosil acid sodium salt (25%) with Fast Blue BB salt (75%) in a 0.2 M Tris-HCl buVer solution (pH 9.3) at room temperature (in the dark).…”
