Purpose The in-vitro environment influences oocyte competence and gene expression in cumulus cells and oocytes. Effects of culturing under non-attachment conditions and varying follicle exposure to FSH were investigated at the mRNA level and on oocyte developmental capacity. Methods Quantitative PCR analysis of Gdf9, Mater, Nmp2 (in oocytes), Lhcgr and Amh (in cumulus cells), and oocyte developmental competence after in vitro follicle culture were evaluated. Results Follicle survival (98.7%) and polar body rate (94%) were similar for all conditions. Estradiol and progesterone production were significantly lower in non-attachment follicles (10-fold and 3-fold, respectively). Under nonattachment conditions, a higher two-cell rate (69.9%) and total blastocyst yield (48.5%) were obtained and, by decreasing FSH levels during culture, Lhcgr transcripts were significantly reduced to levels similar to in-vivo. Levels of oocyte-specific transcripts were not significantly influenced by in-vitro conditions. Conclusion Non-attachment conditions influence follicle steroid secretory capacity and, together with dynamic FSH doses, positively influence cumulus cell gene expression and oocyte developmental competence.

Estrogens play an important role in the growth, differentiation, and function of female reproductive tissues. Estrogen signals through estrogen receptors (ERs), members of the nuclear receptor superfamily. The two major forms, ERalpha and ERbeta, are expressed in the mouse ovary, where ERbeta is predominantly expressed in granulosa cells, and ERalpha in theca cells. In this study, we determined the expression pattern of ER subtypes within mouse follicles cultured from the early preantral stage up to the preovulatory stage and after an ovulatory stimulus in different culture conditions. Immunohistochemical studies performed at different time points of culture revealed that ERbeta was found exclusively in granulosa cell nuclei regardless of follicular growth stage or culture conditions. In contrast, ERalpha was found in oocyte, granulose, and theca cells, and its subcellular localization differed between follicular growth stages and culture conditions. A shift from a predominant cytoplasmic to a predominant nuclear immunolocalization was observed in granulosa cells as follicles reached the antral growth phase, and was postponed in culture conditions with minimal growth factor supplementation. In response to hCG, ERbeta protein levels in luteinized granulosa cells spectacularly declined to undetectable levels, while ERalpha immunostaining again shifted to cytoplasmic regions, but not in theca cells.

Genistein is a plant-derived estrogenic isoflavone commonly found in soy-based products such as soymilk and soy-based dietary supplements for treating menopausal symptoms, for example. Vasopressin is a neurosecretory nonapeptide synthesized primarily in neurons of the hypothalamus and secreted into the bloodstream from the posterior lobe of the pituitary. The endogenous opiate peptide beta-endorphin is synthesized both in neurons of the hypothalamus and in pituitary cells, primarily of the neurointermediate lobe. It has been reported that exposure to 17beta-estradiol or diethylstilbesterol increased the vasopressin content of the hypothalamus, and that estradiol valerate selectively damages hypothalamic beta-endorphin-containing neurons. Since little was known of the potential effects of estrogenic endocrine-disruptor compounds on hypothalamic neuropeptides, we fed Sprague-Dawley fetuses from day 7 in utero until sacrifice at postnatal day 77, with either a control diet (<1 ppm) or an experimental diet containing 25, 250, or 1250 ppm of genistein. We then conducted ELISA assays for hypothalamic content of both beta-endorphin and vasopressin immunoreactivity. Whereas there were no statistically reliable effects of dietary genistein on hypothalamic beta-endorphin content, vasopressin levels were significantly elevated in the 1250-ppm genistein group (p < 0.05). Elevated vasopressin levels may be associated with fluid balance, altered blood pressure, and cardiovascular effects. These data are consistent with the known actions of estradiol and may serve to explain our finding in a previous study that estrogenic endocrine-disruptors such as genistein increased sodium preference in rats exposed through their diet.
