Therapeutic Activity of Pretazettine on Rauscher Leukemia: Combination of Antiviral Activity and Cellular Protein Inhibition

Abstract: Pretazettine hydrochloride (PTZ) has been found to inhibit protein synthesis, without being inhibitory to DNA and RNA, in Rauscher leukemic blood cells in mice for at least 6 h after its administration. With comparison to Virazole and cycloheximide, the specific anti-Rauscher virus activity of PTZ has been demonstrated only in acutely-infected NIH/3T3 cells but not in chronically-infected cells. It is not certain that the inhibitory action of PTZ on reverse transcriptase is contributory to its therapeutic acti… Show more

“…The mechanism of this rever sal effect of PTZ is not known. It may be due to the antiviral [2,3] or antitumor [4. 5] or the immunoenhancing action [personal commun.…”

“…a narcissus alkaloid, was therapeu tically active against Rauscher viral leukemia and spontaneous AKR T cell leukemia [1][2][3]. PTZ is in hibitory to the growth of murine retroviruses in cell cultures and in mice [2,3], PTZ is an inhibitor of viral reverse transcriptase [2] and is cytotoxic due to the specific inhibitory action on cellular protein synthesis [2], and therefore PTZ is also effective against nonviral transplantable tumors such as Ehrlich ascites and Lewis lung carcinoma [4.…”

“…The proba ble active principle has been identified as pretazettine, an unstable alkaloid of the Amaryllidaceae family [5], and a method for the production of the stable hydrochloride of pretazettine (PTZ) for animal use has been established [5], In addition to the re markable antiviral activity against Rauscher and AKR leukemia [3,6], PTZ posesses a moderate or reversible cytotoxicity resulting from the specific inhibitory action on cellular protein synthesis [4,10]. Now we report the therapeutic activity of PTZ against Ehrlich ascites carcinoma and the beneficial adjuvant effect on the thera peutic activity of the individual standard an ticancer drugs in the ABC (adriamycin, BCNU, and cyclophosphamide) regimen on Ehrlich cells.…”
