Therapeutic Efficacy in Experimental Polyarthritis of Viral-Driven Enkephalin Overproduction in Sensory Neurons

Abstract: Rheumatoid arthritis is characterized by erosive inflammation of the joints, new bone proliferation, and ankylosis, leading to severely reduced locomotion and intense chronic pain. In a model of this disease, adjuvant-induced polyarthritis in the rat, neurons involved in pain transmission and control undergo plastic changes, especially at the spinal level. These changes affect notably neurons that contain opioids, such as enkephalins deriving from preproenkephalin A (PA) precursor protein. Using recombinant he… Show more

“…Preclinical studies of chronic pain have used several gene therapy vectors (Cope et al, 2006). Adenoviral, AAV, and HSV vectors have been used to deliver therapeutic genes including β-endorphin (Finegold et al, 1999), proopiomelanocortin (Lu et al, 2002), interleukin 2 (IL-2) (Yao et al, 2003), proenkephalin-A (Braz et al, 2001), IL-10 (Milligan et al, 2005a;Milligan et al, 2005b), glial-derived growth factor (Hao et al, 2003) and neurotrophin-3 (Pradat et al, 2001). …”

“…Attenuated vectors were in fact demonstrated to be highly efficient in driving proenkephalin A (PA) gene expression in dorsal root ganglia (DRG) 75 to deliver genes into monkey eyes, 76 and to the rodent visual system, 77 and to express active nerve growth factor beta subunit (b-NGF) in latently infected DRG. 78 …”
