Therapeutic efficacy of combined vaccination against tumor pericyte-associated antigens DLK1 and DLK2 in mice

Abstract: When compared with vascular cells in normal tissues, pericytes and vascular endothelial cells (VEC) in tumor blood vessels exhibit altered morphology and epigenetic programming that leads to the expression of unique antigens that allow for differential recognition by CD8 C T cells. We have previously shown that the Notch antagonist delta-like homolog 1 (DLK1) is a tumor pericyte-associated antigen expressed in setting of melanoma and a range of carcinomas. In this report, we show that therapeutic vaccination a… Show more

“…In this context, we and others have actively studied therapeutic vascular normalization (VN) as an interventional strategy to promote enhanced immune infiltration and a proinflammatory TME. 3 4 In the VN paradigm originally proposed by Jain, 5 6 provision of antiangiogenic agents at low-moderate (sub-MTD) doses results in improved tumor vascular integrity and perfusion, leading to tissue normoxia, increased stromal production of pro-inflammatory chemokines and augmentation in levels of TIL. 6 7 One class of agents that concomitantly activates robust inflammatory immune responses includes agonists of STING, a cytosolic double-stranded DNA (dsDNA) sensor, which have demonstrated therapeutic potential in early phase clinical trials.…”

“…It also hinted at a role for Dll4 in the maintenance of endothelial barrier integrity. More recently, targeting of a delta-like 1 homolog, a tumor pericyte-associated antigen and Notch antagonist, has led to the development of combined vaccination approaches that lead to reduced vascular permeability and vascular normalization (11) (16). We therefore tested the hypothesis that Dll4 could control the permeability of endothelial barriers in vitro and in vivo.…”

“…Vascularisation is an important component of tumor growth and a target for therapy; the NOTCH antagonist delta-like 1 homologue (DLK1) was previously identified as a tumor pericyte-associated antigen in various carcinomas [ 135 ], also in renal cell carcinomas [ 136 ]. Inhibition of DLK-1 (e.g.…”
