Therapeutic Potential of Natural Products from Terrestrial Plants as TNF-α Antagonist

Abstract: Tumor necrosis factor-alpha (TNF-α) is a multifunctional cytokine produced by monocytes, macrophages, neutrophils, T-cells, mast cells, epithelial cells, osteoblasts and dendritic cells. It can regulate numerous cellular and biological processes such as immune function, cell differentiation, proliferation, apoptosis and energy metabolism. It is also involved in the pathogenesis of multiple chronic inflammatory or autoimmune diseases. The biological activities of TNF-α mediated by two receptors TNFR1 and TNFR2.… Show more

“…This down-regulation of TNF-α seen in the first week may be due to the anti-inflammatory properties of AEGL [17]. Antagonists of Th1-derived TNF-α can be obtained from many natural resources including Stryphnodendron adstringens, Campomanesia lineatifolia, Terminalia glabrescens, and Illicium verum [18].…”

“…Inflammatory cytokine TNF-a acts through NFkB signaling to downregulate CLDN5 expression (70). A multitude of natural product extracts, such as terpenes and phenolic compounds, were shown to be TNF-a inhibitors (71). Asiaticoside, a triterpenoid, enhances endothelial integrity of blood vessels, reducing their permeability through inhibiting TNF-a (72) ( Figure 2).…”

“…The use of medicinal plants and traditional health systems is increasingly emphasized to address global healthcare concerns. [1][2][3][4] Traditional medical practices are considered an essential part of the culture in many emerging countries. Holoptelea integrifolia, or the Indian Elm Tree, is a large deciduous tree found throughout India up to an elevation of 2000 feet.…”
