Therapeutics for advanced hepatocellular carcinoma: Recent advances, current dilemma, and future directions

Abstract: Hepatocellular carcinoma (HCC) is one of the most common malignancies and is a serious threat to people's health worldwide. The prognosis of advanced HCC is dim if left untreated. In the clinic, the treatment options for advanced HCC include surgery, radiotherapy, transcatheter arterial chemoembolization, and so forth. In recent years, molecular targeted therapy and immunotherapy have also made great progress, bringing new hope to patients with advanced HCC. In this study, therapeutic advances, current dilemma… Show more

“…previously, many progresses have been made in understanding the epidemiology, risk factors and molecular profiles of HCC [11]. However, incidence and HCC-specific mortality still continue to increase [12][13][14]. Although some progress has been made in molecular targeted therapy for HCC, the results are still unsatisfactory [15].…”

“…The morbidity and mortality of HCC are particularly higher in Asian countries due to more frequent hepatitis B virus and hepatitis C virus infection [4]. Although surgery, liver transplantation, and radiofrequency ablation have been adopted as curative treatments [5], they are barely available to a considerable portion of HCC patients who are frequently diagnosed in advancedstage [6]. Therefore, it is significantly important to elucidate the molecular mechanism involved in HCC progression, shedding light on new therapeutic targets for HCC patients.…”

“…Despite advances in diagnostic methods, early detection of HC remains difficult [2]. In most patients, HC progresses to the intermediate and advanced stages, for which the 5-year survival rate is <17% [2,3]. Surgery and liver transplantation are considered to be the optimal treatment options for advanced HC; however, only a small proportion of HC patients are able to undergo potentially curative resection [4].…”
