Thermal Inactivation of the Kanagawa Hemolysin of Vibrio parahaemolyticus in Buffer and Shrimp

Abstract: Growth studiesA semipurified thermostable direct hemolysin (Kanagawa hemolysin) was prepared from a clinical isolate of Vibrio parahnemo(vticus (strain 533-72). Preliminary experiments determined that the hemolytic activity and mouse lethality of the hemolysin were inactivated at the same rate, that the rate was linear, and that the pH range of optimum thermal stability was pH 5.5-6.5. When approximately 256 rat erythrocyte units/g of Kanagawa hemolysin were heated at Il5", l20", l25", and 13O"C, D values rang… Show more

“…The D values were calculated from the absolute values of the inverses of the slopes of the linear-regression equations by plotting log numbers of survivors versus time (3) at 300, 350, and 400 MPa at 20°C. The Z values were calculated from the absolute values of the inverses of the slopes of the linearregression equations relating log units (D values) to pressure (4).…”

“…Su manejo y la contaminación cruzada son otro factor importante que afecta la calidad y seguridad del producto (Paris et al, 2005), y finalmente, la reproducción exponencial de la bacteria en lapsos de 10 minutos (Bradshaw et al, 1984). Por esta razón, los moluscos bivalvos son considerados uno de los alimentos más peligrosos para la salud humana (Rippey, 1994).…”

“…Handling and cross-contamination are another important factor affecting the quality and safety of the product (Paris et al, 2005), and finally, the exponential reproduction of the bacteria in 10 minutes lapses (Bradshaw et al, 1984). Thus bivalve molluscs are considered one of the most dangerous foods to human health (Rippey, 1994 (Honda et al, 1988).…”

“…In Tris buffer at pH 7.0, D 120 • C and D 130 • C values of 34 and 13 minutes, respectively, were found for semipurified toxin, whereas in shrimp D 120 • C and D 130 • C values were 21.9 and 10.4 minutes, respectively. 18 Hemolysin was detected when cell counts reached 10 6 /g, and its heat resistance was greater at pH 5.5-6.5 than at 7.0-8.0. 18 The TDH gene (tdh) is chromosomal, and it has been cloned in E. coli.…”
