Three-dimensional culture using human plasma-medium gel with fragmin/protamine microparticles for proliferation of various human cells

Abstract: Fragmin/protamine microparticles (F/P MPs) have been used as carriers for the preservation and activation of cytokines in human plasma (HP)-Dulbecco's modified Eagle's medium (DMEM) gels. This study investigated a three-dimensional (3D) culture system using an HP-DMEM gel with 0.1 mg/mL F/P MPs and 5 ng/mL FGF-2 for the proliferation of human dermal fibroblast cells (DFCs), human microvascular endothelial cells (MVECs) and human coronary smooth muscle cells (SMCs), or 5 ng/mL interleukin (IL)-3/granulocyte-mac… Show more

“…Low concentration of ox-LDL (100 nmol/L) could induce the upregulation of MMP-2 and -9, and miR-29b, inducing the migration of SMCs. Interleukin-3 (IL-3) has been shown to promote cell proliferation and migration in vascular disease (Kishimoto et al 2014). Mir-9 might control the IL-3-induced proliferation and migration of SMCs (Lee et al 2015).…”

“…[ 44 ] 3D cell culture gels induce higher cell proliferation than their 2D culture counterparts for several types of cells, including smooth muscle, endothelial, and fi broblast cells. [ 45 ] This study, however, does not address submicrometer topography, but rather develops a model for 3D cell culture with a fl exible scaffold that has potential to protect the culture during transplantation and during other mechanical stress exposures that require signifi cant material elasticity. An inherent limitation of sub-micrometer laser 3D printing is that the printing time increases as the resolution increases into the sub-micrometer range.…”

“…There are several advantages of developing a platform with plasma as the framework. Being enriched with growth factors and proteins that contribute to the regenerative process, a plasma derived microenvironment potentiates cell proliferation, migration, and differentiation mimicking in vivo conditions, hence enhancing the clinical utility of the model (80)(81)(82)(83). Of note, the use of human plasma as a matrix not only supports cell structure, but also serves as a reservoir for nutrients, growth factors, cytokines, extracellular vesicles, and signaling molecules (84)(85)(86) that help with the development of more relevant human cancer models.…”
