Two species of the genus Sinipta, S. dalmani and S. hectorisperonii, exist in Uruguay, the latter reported here for the first time. Both species have quite similar morphological features and can only be distinguished by slight details, which have led to misidentifications in the past. Specimens from both taxa were studied in order to detect suitable characters to distinguish them and to clarify their taxonomic status, by integrating previously unstudied morphological, biological and molecular characteristics. Detailed structures of the stridulatory file of both sexes were studied and described for the first time, and the sounds produced by both species were studied and described on the basis of recordings made in captivity with an analog recorder. The signal was digitized and analyzed using Avisoft software. Three different types of sound were described for both species: calling song, courtship song and disturbance song. Oscillograms and frequency spectra were provided, and the physical characteristics of the sounds of interest for species identification were established. Finally, molecular studies showed that the two taxa are reciprocally monophyletic groups. The combined results of the analyses suggest that the two studied taxa are distinct species, and point to features suitable for their identification.

Understanding patterns of species diversity relies on accurate taxonomy which can only be achieved by long‐term natural history research and the use of complementary information to establish species boundaries among cryptic taxa. We used DNA barcoding to characterize the ant diversity of Iguazú National Park (INP), a protected area of the Upper Paraná Atlantic Forest ecoregion, located at the southernmost extent of this forest. We assessed ant diversity using both cytochrome c oxidase subunit 1 (COI) sequences and traditional morphological approaches, and compared the results of these two methods. We successfully obtained COI sequences for 312 specimens belonging to 124 species, providing a DNA barcode reference library for nearly 50% of the currently known ant fauna of INP. Our results support a clear barcode gap for all but two species, with a mean intraspecific divergence of 0.72%, and an average congeneric distance of 17.25%. Congruently, the library assembled here was useful for the discrimination of the ants of INP and allowed us to link unidentified males and queens to their worker castes. To detect overlooked diversity, we classified the DNA barcodes into Molecular Operational Taxonomic Units (MOTUs) using three different clustering algorithms, and compared their number and composition to that of reference species identified based on morphology. The MOTU count was always higher than that of reference species regardless of the method, suggesting that the diversity of ants at INP could be between 6% and 10% higher than currently recognized. Lastly, our survey contributed with 78 new barcode clusters to the global DNA barcode reference library, and added 36 new records of ant species for the INP, being 23 of them new citations for Argentina.

The chromosome variability among ant species is remarkable, and the processes generating such variation are still under discussion since polyploidy has been observed in some distinct taxa. The chromosome number of species belonging to the Camponotus, subgenera Myrmothrix and Myrmobrachys, are highly different, whereas, the first subgenus has double the number of chromosomes of the second. In order to test the hypothesis of chromosome number doubling through polyploidy, the genome sizes of Camponotus (Myrmothrix) rufipes, Camponotus (Myrmothrix) renggeri and Camponotus (Myrmobrachys) crassus were estimated by flow cytometry. The chromosome number of specimens from the nests studied was also defined. No significant variation was noted in the genome size among them. The mean haploid genome size value (1C) of workers for the three species was 286.16 Mpb (0.29 pg). The polyploidy hypothesis can be ruled out as an evolutionary step linking the karyotype variations among the three studied species since the genome size of C. crassus with 2n = 20 chromosomes was the same as that of C. rufipes and C. renggeri with 2n = 40. The lack of variation in the amount of DNA between the related species C. rufipes and C. renggeri also demonstrate that flow cytometry is not an adequate approach to distinguish them. Our results highlight the importance of combining distinct methods, DNA quantification, and cytogenetics from the same colony. Understanding the path of chromosome evolution of three species with distinct degrees of relatedness should provide further information in enriching our knowledge about the Minimum Interaction Theory.
