Prostaglandin E1 (PGE1) covalently linked to w-NH2-hexyl-agarose (PGEI-hexyl-agarose) [5,6-3H(N)]PGE1 (74.8 Ci/mmol), and Protosol were purchased from New England Nuclear (1 Ci = 3.7 X 101l becquerels). Cyclic AMP-dependent protein kinase (EC 2.7.1.37) and protein kinase inhibitor were obtained from Sigma. Antisera to prostaglandins were obtained from Miles. PGE1 was kindly provided by J. Pike (Upjohn). All other chemicals were reagent grade.Platelet Preparation. Blood donors had not taken any medication for at least 2 weeks before blood donation. Venous blood was collected through siliconized needles into plastic syringes; coagulation was prevented by mixing 9 vol of blood with 1 vol of trisodium citrate (final concentration, 13 mM). Platelet-rich plasma was obtained by centrifugation of samples for 10 min at 170 X g. The supernatant platelet-rich plasma (3-4 X 108 cells per ml) was collected. Platelet counts were performed in a Coulter Counter model ZB equipped with a 50 ,um aperture tube.Preparation of Insolubilized PGE1. The insolubilized derivative of PGE1 was prepared by coupling the carboxylic moiety of the hormone to the NH2 group of w-NH2-hexylagarose (11). Details of the procedure have been reported (10).Briefly, 14.2 ,gmol of PGE1 was incubated with 5 ml (packed volume) of cw-NH2-hexyl-agarose and 600 mg of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (12) in 10 ml of 50% (vol/vol) The publication costs of this article were defrayed in part by page charge payment.
