“…Genomic Southern blotting was used in order to determine the number of HI' genes in the mouse genome. After 72 h in culture, > 80% of the cells show the characteristic differentiation marker of parietal endoderm such a the cytokeratins 8 and 18, collagen type IV, laminin and a loss of SSEA antigen (for review, see Hogan et al, 1983). wt F9 cell DNA was prepared, cut with different restriction enzymes, blotted onto Genescreen membranes, hybridized to radioactive RNA and autoradiographed.…”

“…Differentiation is expressed by a reprogramming of the transcriptional activity of the cell (Strickland and Mahdavi, 1978;Strickland et al, 1980;Hogan et al, 1983;Wang et al, 1985). This is accompanied by concomitant changes in chromatin structure which are demonstrable by differential sensitivity of selected genes to the action of various nucleases (Croce, 1983).…”

“…Since extraembryonal tissues constitute most of the early conceptus, Kras isoform expression during very early embryogenesis was studied in a range of ES and EC cell lines. Their di erentiation in vitro resembles that of the inner cell mass as it forms parietal or visceral endoderm (Hogan et al, 1983). Di erentiation of ES and EC cell lines over several days following LIF withdrawal was associated with K-rasA induction, and K-rasA was ®rst expressed in the embryo after E9.…”

“…The first indication of differential activity of murine homeo box genes was provided by studying the expression profile of Hox 1.1 (m6) during differentiation of F9 cells in parietal endoderm (Colberg-Poley et al, 1985b). Since F9 cells are nullipotent and thus restricted in their differentiation pathway (Strickland and Mahdavi, 1978;Hogan et al, 1983), mouse embryos were investigated at various stages of development. Specifically, using Northern analyses transient expression of Hox 1.1, Hox 1.2 (Colberg-Poley et al, 1985a,b), Hox 1.3 (M.Fibi, B.Zink, M.Kessel, A.M.Colberg-Poley and P.Gruss, in preparation), Hox 1.4 (Wolgemuth et al, 1986;Rubin et al, 1986;Duboule et al, 1986), Hox 1.5 , Hox 2.1, Hox 2.2 Jackson et al, 1985;Hauser et al, 1985;Krumlauf et al, 1987), and Hox 3.1 (Breier et al, 1986;Awgulewitsch et al, 1986) Hart et al, 1985;Awgulewitsch et al, 1986;Jackson et al, 1985;Krumlauf et al,1987).…”
