Time-evolution of in vivo protein corona onto blood-circulating PEGylated liposomal doxorubicin (DOXIL) nanoparticles

Abstract: Nanoparticles (NPs) are instantly modified once injected in the bloodstream because of their interaction with the blood components. The spontaneous coating of NPs by proteins, once in contact with biological fluids, has been termed the 'protein corona' and it is considered to be a determinant factor for the pharmacological, toxicological and therapeutic profile of NPs. Protein exposure time is thought to greatly influence the composition of protein corona, however the dynamics of protein interactions under rea… Show more

“…1 mL of B or L was mixed with 390 µL of NPs at 4.6 mg Fe mL −1 , corresponding to 1.3 m 2 of NPs mL −1 of serum, close to previously reported values . The PC formation is a dynamic process, which is rapidly changing during the first 2–3 h, as shown in vitro and in vivo . Hence, based on these two reported studies, NPs were incubated for 24 h at 37 °C under agitation either at 1400 or 20 rpm.…”

“…In reality the dispersed particles are hydrated/ solvated which along with its corona are rarely spherical. The composition of the corona -especially the soft corona -is dynamic and fluctuates over time depending on the ionic strength, types of smaller and bigger molecules present in the environment and types of solvents [52,53]. Therefore, DLS provides only an indicative size of the colloid.…”

“…Although relevant, factors defined by NPs are fewer compared to media-dependent factors. Composition of the NPs corona depends on the source of proteins [2931], time [9,32] and temperature [33] of the incubation of NPs in biological fluids. Not all variables are so intuitive; corona composition can also be affected by serum heat inactivation [34], choice of anticoagulant [30], static versus dynamic conditions of the media [35] and even the presence of magnetic field in the case of iron oxide NPs [36].…”
