Time of In Vitro Anther Culture May Moderate Action of Copper and Silver Ions that Affect the Relationship between DNA Methylation Change and the Yield of Barley Green Regenerants

Abstract: Plant anther culture allows for the regeneration of uniform and homozygous double haploids. However, off-type regenerants may appear as a result of so-called tissue culture-induced variation (TCIV). In addition, the presence of Cu2+ and Ag+ ions in the culture medium might influence the number of green plants. The regenerants were obtained via anther cultures of barley under varying Cu2+ and Ag+ ion concentrations in the induction medium during distinct time conditions. DArTseqMet markers were evaluated based … Show more

“…The N6 medium for this study used 6% sucrose, while A1, A2, and L8 used only 1%, 3%, and 5% sucrose, respectively. However, the estimated 2–5% of sucrose levels were good for anther culture in rice [ 103 ]. The results of this study highlighted the significance of the genotype, and media compositions and their interactions on CIF and RGP efficiency (RGPE) in rice.…”

“…In barley anther culture, the two ions may act as mediators of sequence variation that involves different methylation contexts [ 74 ]. They may also affect green plant regeneration [ 75 ] and somewhat differently act in somatic embryogenesis [ 76 ]. Thus, playing with the concentration of copper and silver ions in the induction medium may influence the variation level.…”

“…This method takes advantage of the properties of Acc 65I and Kpn I isoschizomers, which recognize the same restriction site but differ in their sensitivity toward the DNA methylation pattern at the restriction site ( Bednarek et al, 2007 ; Mikula et al, 2011b ; Coronel et al, 2018 ). We recently used metAFLP to analyze DNA methylation patterns in barley regenerants derived via anther culture ( Bednarek and Orłowska, 2020a , c ; Bednarek et al, 2020 ).…”
