Tirofiban combined with urokinase selective intra-arterial thrombolysis for the treatment of middle cerebral artery occlusion

Abstract: Abstract. The aims of the present study were to establish a model of embolic stroke in rabbits and to evaluate the efficacy and safety of intra-arterially administered tirofiban combined with urokinase thrombolysis. The middle cerebral artery occlusion model (MCAO) of embolic stroke was established in New Zealand rabbits via an autologous clot. The model rabbits were allocated at random into four groups: Tirofiban group (T group), urokinase group (UK group), tirofiban and urokinase group (T + UK group) and the… Show more

“…Behavioral tests can be performed on pigs, 31 dogs, 152 rabbits, 153 NHPs, 154 and sheep. 30 However, only a small proportion of the studies used behavioral endpoints (6%, 20/334; Figure 4(a)).…”

“…If IAT was combined with other therapeutic strategies (e.g., mechanical thrombectomy, intravenous thrombolysis or tirofiban), the dosage could be reduced to 100,000-200,000 IU [24,25]. The dose of urokinase used in this study was less than that for IAT in large artery occlusion (mostly 500,000-1,000,000 IU) and more than that in combined therapies when it comes to the non-large artery occlusion.…”

“…Our case was only treated through the intra-arterial method, achieving a good result. Tirofiban is usually used as an addition to endovascular treatment, to prevent the reocclusion of the recanalized arteries [ 12 , 13 ]. Our observations indicate that the potential use of tirofiban as a thromblytic agent, and suggest further study to assess the role of intra-arterial tirofiban as a single treatment for acute ischemic stroke.…”
