Tissue clearing technique: Recent progress and biomedical applications

Abstract: Systematic investigation of the morphology and structure of the organism at single-cell resolution is essential for elucidating the associated mechanisms in health and disease. However, extracting detailed information with a global perspective from an intact biological specimen has been a fundamental challenge in biomedicine, because biological tissues are mostly opaque, which hinders light penetration into deep tissue and restricts imaging depth and contrast (Zhu et al., 2013). Conventional histological secti… Show more

“…The use of tissue clearing in human health—and more particularly for respiratory diseases—emerges and has already been reported for the analysis of lung organoids or lung biopsies from human patients [ 39 , 57 ]. In combination with biochemistry and histology, tissue clearing and 3D imaging will contribute to personalized medicine [ 58 ].…”

“…However, the main focus of tissue clearing methods is in neuroscience and brain development. The majority of original papers and reviews in tissue clearing as well as 3D brain CUBIC-atlas come from this field (Murakami et al, 2018;Tian et al, 2021;Ueda et al, 2020;Vigouroux et al, 2017). However, the number of publications discussing tissue clearing methods on heart is limited (Kolesova et al, 2016;Nehrhoff et al, 2016;Yokoyama et al, 2017), even though the field of heart and vasculature development has its avid audience.…”
