Tissue Culture Independent Agrobacterium tumefaciens Mediated In Planta Transformation Method for Tropical Maize (Zea mays.L)

Abstract: Tropical maize is recalcitrant to tissue culture regeneration because of its poor response to in vitro regeneration after transformation. In this context, the present study has developed the tissue culture independent in planta transformation protocol for tropical maize by transferring plumular meristem cells of germinating seeds of tropical maize genotype through Agrobacterium tumefaciens infection. The protocol was developed by using Agrobacterium strain EHA105 containing vector pCAM-BIA3301 carrying cry1Ab,… Show more

“…In some protocols, a selection step under in vivo conditions (e.g. maize [133]) or in vitro conditions using soilrite as a medium (e.g. roselle/Hibiscus sabdariffa [134]) has been added after inoculation to select the best-performing T 0 chimeric plants.…”

“…However, the transformation frequency was reported to be quite low and the screening of the transformants by PCR analysis was time-consuming. Abhishek et al (2014b) developed a tissue culture independent protocol for in planta transformation in tropical maize by using plumular meristems of germinating seeds as explants and transforming them using Agrobacterium approach.…”

“…The establishment of a sesame regeneration system has been problematic, and therefore, sesame was labeled a recalcitrant plant, 13 , 14 with a minimal-successful rate of genetic modification. 14 , 16–18 , 19 , 20 In the present work, we report a new, more straightforward, non-tissue culture-based method for rapid and stable transformation of sesame seeds using the ‘imbibed seed co-cultivation method’ (ISCM).…”
