Tissue Distribution of Doxycycline in Animal Models of Tuberculosis

Abstract: Doxycycline, an FDA-approved tetracycline, is used in tuberculosis in vivo models for the temporal control of mycobacterial gene expression. In these models, animals are infected with recombinant Mycobacterium tuberculosis carrying genes of interest under transcriptional control of the doxycycline-responsive TetR-tetO unit. To minimize fluctuations of plasma levels, doxycycline is usually administered in the diet. However, tissue penetration studies to identify the minimum doxycycline content in food achieving… Show more

“…The ratios C max /EC 50 and C max /EC 90 in plasma seem low to reach effective concentrations to inhibit SARS-CoV-2 in humans. However, in lungs, doxycycline were 2-to 4-fold higher than in plasma [39,40]. A daily oral uptake of 100 or 200 mg of doxycycline led to a C max value from 3.4 to 20 µg/g of lungs.…”

“…The mice were fed doxycycline chow starting 3 days before infection to repress sspB expression in the mutants upon infection, and allow them to grow normally. At 3 weeks post-infection, doxycycline was withdrawn to initiate protein degradation, and two weeks of RIF treatment was administered (Fig7B) (58). The antibiotic regimen effectively decreased the total number of lung CFU (Fig7C) .…”

“…Lung tissue was collected from female New Zealand white (NZW) rabbits (Millbrook Farm, Concord, MA), for the preparation of the tissue spotting and tissue mimetic models. Rabbits were maintained under specific pathogen‐free conditions with access to food and water ad libitum, as previously described 67 …”
