Tissue-targeted R-spondin mimetics for liver regeneration

Abstract: R-spondin (RSpo) proteins amplify Wnt signaling and stimulate regeneration in a variety of tissues. to repair tissue in a tissue-specific manner, tissue-targeted RSPO mimetic molecules are desired. Here, we mutated RSPO (RSPO2 F105R/F109A) to eliminate LGR binding while preserving ZNRF3/RNF43 binding and targeted the mutated RSPO to a liver specific receptor, ASGR1. The resulting bi-specific molecule (αASGR1-RSPO2-RA) enhanced Wnt signaling effectively in vitro, and its activity was limited to ASGR1 expressing… Show more

“…Here, we describe development of L6-F4-2, a selective tetravalent potent surrogate of the FZD 4 :LRP6 receptor complex. L6-F4-2 does not bind FZD 1-3 or FZD 5-10 ECDs but promotes proximity of FZD 4 to LRP6 coreceptors, consistent with our prior synthesis of bioengineered WNT surrogates targeting other FZD family members 3538,40,64 . Compared to Norrin and the recently reported FZD 4 -selective WNT surrogate F4L5.13 28 , L6-F4-2 exhibits approximately 100 times higher FZD 4 affinity and potency of WNT/β-catenin signaling activation in vitro.…”

“…While other agonists of Wnt signaling have been generated, our molecule directly engages Wnt receptor and co-receptor and hence is likely more potent and specific. 6 , 49 A direct comparison with such agents is not currently feasible but may be necessary in the future. FL6.13 has been shown to activate Wnt signaling at other sites, including eye and intestines, based on the method of delivery.…”

“…Western blot analysis was performed as previously reported, 30 with minor modifications. In brief, HRMECs were treated with 10-nM SZN-413-p 24 hours before harvesting for cell lysis.…”
