Tissue transglutaminase as a central mediator in inflammation-induced progression of breast cancer

Abstract: TGM2 is a stress-responsive gene that encodes a multifunctional and structurally complex protein called tissue transglutaminase (abbreviated as TG2 or tTG). TGM2 expression is frequently upregulated during inflammation and wounding. Emerging evidence indicates that TGM2 expression is aberrantly upregulated in multiple cancer cell types, particularly those selected for resistance to chemotherapy and radiation therapy and those isolated from metastatic sites. It is becoming increasingly evident that chronic expr… Show more

“…Although TG2 has been shown to drive pro-inflammatory NFκB signaling (34, 3844), our studies show that NFκB signaling is not relevant in ECS cells (4, 5). Thus, how TG2 targets downstream signaling mechanisms to maintain cancer stem cell properties is not well understood (10).…”

“…TG2 is an important pro-inflammatory regulator in cancer cells (9, 3436). Our studies show that TG2 level is markedly elevated in ECS cells as compared to non-stem cancer cells, and that TG2 is required for ECS cell survival, invasion, EMT, migration and tumor formation (4, 4, 5).…”

“…Based on the demonstrated importance of TG2 in several epithelial cancers (3337), we examined expression of TG2 in skin cancer. Our studies show that TG2 levels are markedly elevated in ECS cells as compared to bulk cultures of cancer cells.…”

“…The fact that these markers are highly expressed in infiltrating breast carcinoma [14, 15] prompted us to investigate their expression in CIS to better understand their functional role in the progression to the infiltrating phenotype. We also considered stromal cells surrounding the tumor following recent evidence of the role of the microenvironment in cancer progression.…”
