TLR/WNT: A Novel Relationship in Immunomodulation of Lung Cancer

Abstract: The most frequent cause of death by cancer worldwide is lung cancer, and the 5-year survival rate is still very poor for patients with advanced stage. Understanding the crosstalk between the signaling pathways that are involved in disease, especially in metastasis, is crucial to developing new targeted therapies. Toll-like receptors (TLRs) are master regulators of the immune responses, and their dysregulation in lung cancer is linked to immune escape and promotes tumor malignancy by facilitating angiogenesis a… Show more

“…TLR signaling pathways are involved in promoting cancer cell proliferation, invasion, and metastasis [8,28,29]. Activation of TLRs in NSCLC cells leads to upregulation of pro-inflammatory cytokines and chemokines (TNF-α or CCL2), contributing to the immunosuppressive nature of the tumor microenvironment [30][31][32][33][34][35][36]. TLRs (especially TLR4) have been shown to influence immune evasion mechanisms in NSCLC.…”

“…Generally, WNT5A is an oncogenic protein involved in invasion and metastasis processes that increases its expression in human bronchial epithelial cells after exposure to cigarette smoke ( 92 , 93 ). At the same time, tobacco smoke could induce the expression of TLRs in macrophages becoming both a very potent inducer of lung cancer and a trigger factor contributing to the unbalancing of Wnt/TLR ( 94 , 95 ). TLRs have been discovered to be potential prognostic markers for lung cancer.…”

“…The Wnt ligands in the TME could interact and activate the TLR pathways. These TLR signalling mechanisms enhance the secretion of cytokines [20]. Inflammatory cytokines, such as interleukins and tumour necrosis factor (TNF), can create a pro-inflammatory environment that supports cancer development.…”
