TNFα and Immune Checkpoint Inhibition: Friend or Foe for Lung Cancer?

Abstract: Tumor necrosis factor-alpha (TNFα) can bind two distinct receptors (TNFR1/2). The transmembrane form (tmTNFα) preferentially binds to TNFR2. Upon tmTNFα cleavage by the TNF-alpha-converting enzyme (TACE), its soluble (sTNFα) form is released with higher affinity for TNFR1. This assortment empowers TNFα with a plethora of opposing roles in the processes of tumor cell survival (and apoptosis) and anti-tumor immune stimulation (and suppression), in addition to angiogenesis and metastases. Its functions and biomar… Show more

“…Article https://doi.org/10.1038/s41467-022-33052-y on angiogenesis 70 , as its promoting/inhitibory effect on angiogenesis is context-dependent 71 . Despite cDCs not being enriched in the tumor setting in our dataset (Supplementary Data 4, 5), these results nevertheless highlight the putative importance of further investigation into cDC-EC crosstalk in light of tumor angiogenesis in BC.…”

“…Benoot et al. evaluated the numerous contradictory findings of TNFα application in lung cancer ( 138 ), bringing to our awareness the varied functions of various TNF family members and the positive impacts of TNF signaling. Modern genomic, transcriptomic, and proteomic techniques are useful for identifying signaling events and molecules in signal transduction ( 139 , 140 ).…”

“…TNFRSF members can stimulate T-cell responsiveness and define niches for T cell memory ( 21 , 22 ). Several TNFRSF members have been identified as emerging cancer immunotherapy targets ( 23 25 ). Unfortunately, the expression profile and clinical relevance of the TNF family in SCLC remain poorly understood.…”
