Tolerability of  -Blockade With Doxazosin as a Therapeutic Option for Symptomatic Benign Prostatic Hyperplasia in the Elderly Patient: A Pooled Analysis of Seven Double-Blind, Placebo-Controlled Studies

Abstract: Background. Benign prostatic hyperplasia (BPH) is a common disorder in males, and its incidence increases with age. Safety results from trials with doxazosin therapy in elderly patients were collated and described.

“…However, several findings question this conclusion. Doxazosin and terazosin lower blood pressure to a greater extent in hypertensive compared with normotensive patients with LUTS, but their incidence of adverse effects appears to be similar [46]. A slow-release formulation of doxazosin similarly was effective in blood pressure and LUTS reduction as the standard formulation, but had an improved tolerability [47].…”

“…The analyzed pooled safety data from seven completed multicenter, double-blind, placebo-controlled studies of doxazosin treatment in patients with BPH, after stratification by age and blood pressure status, 36 included 315 normotensive patients aged o65 y and 320 normotensive patients aged X65 y. Neither of these normotensive age groups showed clinically important reductions in mean blood pressure during treatment with doxazosin.…”

“…In that study [60], alfuzosin produced a more marked blood pressure reduction than tamsulosin, but both drugs had a very similar adverse event pro®le, which was similar to that of placebo. Whereas doxazosin [61,62] and terazosin [63] lower blood pressure to a greater extent in hypertensive than in normotensive patients with LUTS, their incidence of side-effects is similar in both normotensive and hypertensive patients [64]. More recently, both the slow-release formulations of doxazosin and alfuzosin have been shown to be similarly effective in both blood pressure and LUTS reduction as the standard formulation, but with improved tolerability [65,66 . ].…”
