Tomato R2R3-MYB Proteins SlANT1 and SlAN2: Same Protein Activity, Different Roles

Abstract: Anthocyanins are water-soluble polyphenolic compounds with a high nutraceutical value. Despite the fact that cultivated tomato varieties do not accumulate anthocyanins in the fruit, the biosynthetic pathway can be activated in the vegetative organs by several environmental stimuli. Little is known about the molecular mechanisms regulating anthocyanin synthesis in tomato. Here, we carried out a molecular and functional characterization of two genes, SlAN2 and SlANT1, encoding two R2R3-MYB transcription factors.… Show more

“…Through mining the WGBS datasets, we also identified an MYB TF gene SlAN2 which is involved in regulating expression of genes required for anthocyanidin biosynthesis (Povero et al, 2010;Kiferle et al, 2015). Compared to AC and rin, the DMR in the SlAN2 gene promoter was almost completely free of DNA methylation in normal Cnr fruit.…”

“…7A-D), genes related to pigment formation and color, were considerably higher at the fully ripe stage. These increments were positively correlated with the expression of R2R3-Myb [30,31] (Fig. 7 E), the plant transcrip- tion factor which is highly correlated to the anthocyanin accumulation, while the expression of translation elongation factor 1, used as a housekeeping gene, was nearly equal during the all three stages (Fig.…”

“…For instance, in tomato, MYB transcription factor encoded by two paralogs of tomato's anthocyanin genes; SlANT1 and SlAN2 which has the potential of regulating fruit's anthocyanin pigmentation and color [99]. These two genes induce anthocyanin synthesis in its fruit when their expression is driven by the 35S promoter (35S: SlANT1 and 35S: SlAN2) and both of them have shown the high expression levels of SlDFR which has capable of encoding a key enzyme in anthocyanin biosynthetic pathway.…”
