Topical 1% propranolol cream promotes cutaneous wound healing in spontaneously diabetic mice

Abstract: Vascular endothelial growth factor (VEGF) expression was significantly lower in the experimental vs. control group whereas NG2 proteoglycan was increased throughout the study. However, matrix metallopeptidase (MMP)-9 expression was at first significantly higher in the experimental vs. control group then the MMP-9 protein level in the control group increased and surpassed that in the experimental group. In conclusion, intralesional administration of 1% propranolol cream promotes reepithelialization and regulate… Show more

“…It has been reported that β- adrenergic antagonists may alter the activity of MMPs. For example, the topical application of propranolol in wounds of diabetic mice induces the expression of MMP-9, which stimulates the reepithelialization in the early wound-healing stages [38]. Our results suggest that the inhibition of dopamine-induced MMP-9 activity in keratinocytes would be mediated, at least in part, by the activation of β-ARs.…”

“…Unbalanced expression of VEGF leads to vasodilation and increased vascular permeability to inflammatory cells, possibly perpetuating the skin damage in psoriasis and PF. Therefore, regulation of VEGF and endoglin expression in patients with PF using anti‐VEGF therapy such as topical propranolol may enable adequate healing and disease remission. Further studies are necessary to evaluate the effects of these drugs.…”

“…6 b-blockers can also reduce the circulating and tissue levels of VEGF, thus enabling the formation of mature vessels to achieve adequate healing. 7 As the expression of all types of ADRB was observed in the epidermis of the majority of skin fragments from patients with PF, it is reasonable to believe that adding propranolol to the treatment of PF lesions in patients with dysregulated expression of VEGF and endoglin could promote adequate angiogenesis and keratinocyte migration, thus improving re-epithelization. Our group is developing additional studies with a larger sample size to confirm this hypothesis.…”
