Topology of the Type IIa Na+/Pi Cotransporter

Abstract: Abstract. The type IIa Na + /P i cotransporter (NaPiIIa) plays a key role in the reabsorption of inorganic phosphate (P i ) in the renal proximal tubule. The rat NaPi-IIa isoform is a protein of 637 residues for which different algorithms predict 8-12 transmembrane domains (TMDs). Epitope tagging experiments demonstrated that both the N and the C termini of NaPi-IIa are located intracellularly. Site-directed mutagenesis revealed two N-glycosylation sites in a large putative extracellular loop. Results from str… Show more

“…The proposed topology of the eukaryotic isoforms comprises 12 α-helices (designated as transmembrane domains (TMDs)) (Fig 5A). This model combines predictions from biochemical and biophysical studies including epitope labeling (Lambert, Traebert et al 1999), cysteine scanning mutagenesis (see section IV.A.2) and in vitro glycosylation assays (Radanovic, Gisler et al 2006). The C-terminal region is important for targeting, hormonal regulation and proteinprotein interactions.…”

“…Secondary topology of SLC34 family proteins. Topology is based on the rat NaPi-IIa sequence and predicted according to (81). One essential cysteine bridge (black, dashed line) links the N-and C-terminal halves of the protein and there are two N-glycosylation sites in the large extracellular loop.…”

“…1B). Topology predictions indicate that these regions include two membrane spanning domains that we have postulated to form re-entrant domains localized between TMDs 2 & 5 and TMDs 7 &10 respectively [38,33]. From functional studies using Cys substitutions, we proposed that these putative re-entrant domains may associate to form the P i transport pathway and, by implication, are also involved in the translocation of the co-substrate (Na + ) in the functionally associated leak mode that operates in the absence of external P i [3,20].…”

“…Based on cysteine (Cys) scanning mutagenesis and in vitro transcription/translation assays, a topology model was proposed for NaPi-IIa that is most likely also valid for NaPi-IIb and NaPi-IIc [33]. This model predicts 12 transmembrane-spanning domains (TMDs), with intracellular NH 2 and COOH termini (Fig.…”
