Toward Realistic Dosimetry In Vitro: Determining Effective Concentrations of Test Substances in Cell Culture and Their Prediction by an In Silico Mass Balance Model

Abstract: Nominal concentrations (C Nom ) in cell culture media are routinely used to define concentration−effect relationships in the in vitro toxicology. The actual concentration in the medium (C Medium ) can be affected by adsorption processes, evaporation, or degradation of chemicals. Therefore, we measured the total and free concentration of 12 chemicals, covering a wide range of lipophilicity (log K OW −0.07−6.84), in the culture medium (C Medium ) and cells (C Cell ) after incubation with Balb/c 3T3 cells for up … Show more

“…To improve the comparison between different cell systems, it is essential to determine the concentration that is actually available to be taken up by the cells in the in vitro system. In the case of lipophilic compounds such as perhexiline, in vitro system components such as Matrigel or medium components can decrease this available concentration [ 56 , 57 ]. In the case of bupropion (CYP2B6 activity), abiotic degradation of the compound was observed, especially in the HL-ICOs.…”

“…In vitro methods for determining air to blood partition coefficients include artificial membrane models, liquid–liquid extraction, Parallel Artificial Membrane Permeability Assay (PAMPA), cell culture models, , and Selected Ion Flow Tube Mass Spectrometry (SIFT-MS). , Additionally, chromatographic techniques, like gas chromatography (GC) and liquid chromatography (LC), can also be used, with two-dimensional gas chromatography (GC×GC) being a powerful tool, offering enhanced resolution for volatile compound analysis and their complex mixtures. , Despite avoiding live animal testing and reducing ethical concerns, in vitro testing has drawbacks like being costly, laborious, and complex. , …”

“…It is obvious that the quantification of Colc content in pharmaceutical and biological samples is very important in order to monitor the treatment of patients and perform pharmacological studies. Several analytical methods have been reported for Colc quantification, such as high-performance liquid chromatography (HPLC), mass spectroscopy (MS), spectrophotometry, HPLC-MS, and HPLC-UV [ 11 , 12 , 13 , 14 , 15 ]. Despite the advantages of these techniques, they require time-consuming sample preparation procedures and expensive instrumentation.…”
