Towards pharmacological intervention in inositol pyrophosphate signalling

Abstract: To help define the molecular basis of cellular signalling cascades, and their biological functions, there is considerable value in utilizing a high-quality chemical ‘probe’ that has a well-defined interaction with a specific cellular protein. Such reagents include inhibitors of protein kinases and small molecule kinases, as well as mimics or antagonists of intracellular signals. The purpose of this review is to consider recent progress and promising future directions for the development of novel molecules that… Show more

“…Inositol pyrophosphates regulate diverse cellular processes [19] , [20] , [21] , [22] , [23] . The most characterized inositol pyrophosphate in mammals, is 5-IP7, which is synthesized by a family of three inositol hexakisphosphate kinase enzymes, of which IP6K1 is the major isoform [19] , [24] , [25] .…”

“…Accordingly, TNP reduces 5-IP7 levels in various cells including adipocytes [19] , [32] , [45] , [54] , [56] , [57] . Furthermore, 5-IP7 is the precursor of PPIP5K mediated production of the higher inositol pyrophosphate 1,5-IP8 [22] , [55] . Accordingly, TNP reduces both 5-IP7 and 1,5-IP8 levels in NIH3T3 cells [58] .…”

“…Inositol phosphate kinases (IP3K, IPMK, ITPK1, IP5K, IP6K and PPIP5K) perform numerous biological processes through their participation in a carefully-regulated, metabolic network that converts phospholipase C-derived Ins(1,4,5)P 3 into an array of more highly phosphorylated cell-signaling molecules [ 1 3 ]. Among these metabolites, considerable attention is currently being focused upon the inositol pyrophosphates (PP-InsPs), the distinguishing feature of which is the possession of ‘high-energy’ diphosphate groups at the 1- and/or 5-positions of the six carbons that comprise the inositol ring [ 3 , 4 ]. Multiple and diverse cellular activities have been attributed to the PP-InsPs, but an over-arching hypothesis views them as acting as an interface between energy metabolism and cell-signaling [ 3 , 5 , 6 ].…”

“…Moreover, IP6K1-KO mice are lean due to enhanced energy expenditure as energy intake is unaltered in these mice (Chakraborty A, 2010). Therefore, IP6K1 is a target in obesity/T2D (Boucher J, 2014; Chakraborty A, 2011; Mackenzie RW, 2014; Shears, 2016). However, the precise mechanism/s by which IP6K1 regulates lipid metabolism in not known.…”
