Toxicity and Surface Modification of Dendrimers: A Critical Review

Abstract: : As compared to other nano polymers, dendrimers have novel three dimensional, synthetic hyperbranched, nano-polymeric structures. The characteristic of these supramolecular dendritic structures has a high degree of significant surface as well as core functionality in the transportation of drugs for targeted therapy, specifically in host-guest response, gene transfer therapy and imaging of biological systems. However, there are conflicting shreds of evidence regarding biological safety and dendrimers toxicity … Show more

“…The potency of viral inhibition reported in these studies was not as significant as ours. Moreover, LNPs and positive charge dendrimer have been found to be capable of inducing immune or cell toxicity (Kedmi et al , 2010 ; Kharwade et al , 2021 ), which might limit the safety window for dosing. These properties also increase the difficulty and cost of industrial production.…”

“…The cytotoxic activity, hemolytic properties, cellular uptake, or ability to transport and release different cargo compounds of many dendrimers have already been studied in vitro [ 9 , 71 ]. However, studies in isolated groups of cells do not accurately model the difficulties nanoparticles must overcome to reach their targets in vivo.…”

“…However, there are specific concerns that require further attention. Firstly, unmodified PAMAM materials with positively charged surfaces are generally considered toxic [74]. Secondly, smaller PAMAMs can be easily eliminated by the kidneys; in contrast, larger ones tend to be recognized by the reticuloendothelial system (RES) [75].…”
