Tracer-derived total and folate-dependent homocysteine remethylation and synthesis rates in humans indicate that serine is the main one-carbon donor

Abstract: , respectively). However, the rate of overall homocysteine remethylation (ϳ8 mol⅐kg Ϫ1 ⅐h Ϫ1) was twice that of previous reports, which suggests a larger role for homocysteine remethylation in methionine metabolism than previously thought. By use of estimates of intracellular [3-13 C]serine enrichment based on a conservative correction of plasma [3-13 C]serine enrichment, serine was calculated to contribute ϳ100% of the methyl groups used for total body homocysteine remethylation under the conditions of this p… Show more

“…Plasma isotopic enrichments of [1-13 C]methionine, [methyl-2 H 3 ]methionine, and [1-13 C]homocysteine were quantified on the heptafluorobutyric anhydride (HFBA) derivatives by GC-MS by using a modification of the methods described (37,42). Blood 13 CO 2 enrichment was determined by gas isotope ratio mass spectrometry (26).…”

“…Blood 13 CO 2 enrichment was determined by gas isotope ratio mass spectrometry (26). Plasma homocysteine was determined by isotope dilution using D 8 -homocystine (37,42). Briefly, plasma was acidified with 10% trichloroacetic acid, amino acids were separated by cationexchange (AG 50W , and the concentration of homocysteine was calculated by isotope dilution as described (37).…”

“…However, the mechanism(s) through which elevated circulating levels of Hcy promote vascular disease and thrombosis is still unclear (9). Hcy has two primary fates: conversion through a reaction catalyzed by 5,10 methylenetetrahydrofolate reductase (MTHFR) into L-methionine or conversion to L-cysteine (L-Cys) via a transulfuration pathway (10,11). The transulfuration pathway relies upon cystathionine β-synthase (CBS) to transform Hcy in cysthathionine, which is converted by cystathionine γ-lyase (CSE) into L-Cys.…”
