Track-etched membrane microplate and smartphone immunosensing for SARS-CoV-2 neutralizing antibody

Abstract: The level of neutralizing antibody (NAb) to SARS-CoV-2 could be used to evaluate the acquired protective immunity of COVID-19 patients or vaccinees. Here we develop a track-etched microporous membrane filtration microplate (TEM) and optical fibers transmitted immunosensing smartphone platform (TEMFIS) based surrogate virus neutralization test (TEMFIS-sVNT) for rapid one-step testing of NAb to SARS-CoV-2. Coefficient variation (CV) of intra-assay and inter-assay precisions of TEMFIS-sVNT varied below 9% or 14%,… Show more

“…Finding alternative methods without cell and virus culture operation is of interest to obtain reliable NAb information for in vitro assays, which are simple, fast, high-throughput, and commercially available. Wang et al [ 38 ] designed a track-etched membrane microplate and portable immunosensing smartphone reader platform (TEMFIS) for simply and quantitatively detecting NAb against SARS-CoV-2, and it exhibited good performance in detecting serum/plasma samples from COVID-19 patients and vaccines. In comparison with the virus neutralization testing and ELISA, the detection by TEMFIS is simple and can be completed within 45 min.…”

“…To achieve quantitative measurement while maintaining a simple and user-friendly interface, attempts have been made to conduct immunoassays in miniaturized devices (28) using principles such as ELISA (29)(30)(31), electrochemistry (32,33), surface plasmon resonance (34), and fluorescence (35)(36)(37)(38). However, signal quantification requires additional detection modules, e.g., optical fibers, light source, amperemeters, or photodetectors, for measuring optical absorbance or fluorescence intensity.…”

“…Despite being rapid, the LFA-based screening kits are usually of lower sensitivity and specificity when compared to the qRT-PCR test [ 19 , 20 ]. Methods based on surface-enhanced Raman Spectroscopy (SERS), field-effect transistor (FET), electroluminescence, localized surface plasmon resonance (LSPR) and electrochemical sensing have been demonstrated in the literature [ [21] , [22] , [23] , [24] , [25] , [26] , [27] ]. Despite the high sensitivity of these methods, their complexity, high cost, and low reproducibility, hinder their translation to POC use.…”
