Tracking Mercury in Individual Tetrahymena Using a Capillary Single-Cell Inductively Coupled Plasma Mass Spectrometry Online System

Abstract: As a kind of unicellular eukaryotic protozoa, Tetrahymena is located at the bottom of the aquatic food webs and plays an essential role in the bioaccumulation of mercury (Hg). To track Hg in individual Tetrahymena, a capillary single-cell inductively coupled plasma mass spectrometry (ICPMS) online system was developed. The experimental and instrumental conditions were optimized to ensure the signal detected was the Hg uptake in individual Tetrahymena. Moreover, a quantitative method was established and validat… Show more

“…The iterative algorithm data processing strategy is widely used for MS data processing in single-cell analysis. , The iterative calculation for the whole data set through the 3 times standard deviation (3σ) finds the signal threshold, and all bursts with an intensity higher than this threshold are collected and considered as the nanoparticle or cellular events. This approach is convenient for data processing in the situation that one data point represents a single-cell signal (single-cell event).…”

“…Eighteen (18) fish samples from sampling sites S 8 and S 9 in the Asunafo South district of the Tano Basin were studied between November 2016 and October 2017. e levels of seven trace metals mercury (Hg), cadmium (Cd), lead (Pb), copper (Cu), arsenic (As), zinc (Zn), and chromium (Cr) in the muscles of fish samples from River Tano were studied. e selection of these metals was based on their toxicity, medical importance, bioaccumulation, and persistence [28]. e study area was mapped and georeferenced with the aid of a hand-held Garmin 62SC Geographical Positioning System appliance [12].…”

“…Ingestion of fish polluted with high levels of Hg, As, and Cr is associated with health problems, including kidney, liver, and skin cancers [1,12,26]. For example, the Minamata disease in Japan in the 60s was attributed to methylmercury (MeHg) exposure through seafood consumption [28].…”

“…In contrast to the soft ion sources, ICP is a high-temperature ion source in which all the chemical bonds in a sample (e.g., a cell) are broken and the generated elemental isotopes are ionized efficiently, thus making ICP–MS a powerful method for ultratrace elemental analysis . Trace elements, metal drugs, , and metal NPs , in single-cell suspensions have been successfully analyzed with ICP–MS. By element-tagged antibody labeling of single cells, ICP–MS equipped with a time-of-flight mass analyzer enables the simultaneous determination of more than 30 markers (antigens) in a single cell in suspension …”
