Trajectories of peripheral interleukin-6, structure of the hippocampus, and cognitive impairment over 14 years in older adults

Abstract: We aimed to investigate if trajectory components (baseline level, slope and variability) of peripheral IL-6 over time were related to cognitive impairment and smaller hippocampal volume, and if hippocampal volume explained the associations between IL-6 and cognitive impairment. Multivariable regression models were used to test the association between IL-6 trajectory components with change in neuroimaging measures of the hippocampus, and with cognitive impairment among 135 older adults (70–79 years at baseline)… Show more

“…Alongside which, our selected measure of physical fatigability was only ascertained at age 68. However, there are strengths of using IL-6 if only a limited number of markers are available [56] and we were able to assess the potential for reverse causality by adjusting for an earlier measure of fatigue. While BMI has been measured at multiple ages in the NSHD, we focused on age 60–64 so that we could investigate inter-relationships with inflammation.…”

“…Cognitive status was clinically adjudicated among participants at the Pittsburgh site during the 2010–2012 site visit from data collected at that and prior visits, as described elsewhere [19]. Participants were characterized as having normal cognition, MCI, or dementia.…”

“…Cognitive status was clinically adjudicated on Health ABC participants who were seen at the year 14 site visit in 2011 to 2012 from data collected at the year 14 and prior visits, as described elsewhere. 13 Because of the restricted sample size, participants were categorized as cognitively normal or cognitively impaired (MCI or dementia) for these analyses.…”
