Transcription factors COUP-TFI and COUP-TFII are required for the production of granule cells in the mouse olfactory bulb

Abstract: Neural stem cells (NSCs) persist in the adult mammalian subventricular zone (SVZ) of the lateral ventricle. Primary NSCs generate rapidly dividing intermediate progenitor cells, which in turn generate neuroblasts that migrate along the rostral migratory stream (RMS) to the olfactory bulb (OB). Here, we have examined the role of the COUP-TFI and COUP-TFII orphan nuclear receptor transcription factors in mouse OB interneuron development. We observed that COUP-TFI is expressed in a gradient of low rostral to high… Show more

“…ASCL1 was expressed in the progenitors in the SVZ, RMS and the core of the OB in the adult mouse. Staining results of brains sections produced a pattern of ASCL1 immunoreactivity that was identical with previous descriptions (Li et al, ; Xu et al, ; Zhou et al, ).RRID: AB_10709354 Goat anti‐SP8 (Santa Cruz Biotechnology Cat# sc‐104661, , Dilutions 1:1,000, polyclonal. “…S13). Among them, several known neuronal TFs, like NEUROD1, NEUROG2, POU3F2, or MYT1L, reconstitute <20% of the P19 program, while other "early" factors, like ASCL1 (Huang et al 2012(Huang et al , 2015, NR2F2 (Zhou et al 2015), or NR4A2 (Park et al 2006) reconstitute >60% (Fig. 6B).…”

“…In addition, as progenitors divide while migrating, they are continuously exposed to signalling molecules along their paths. COUP-TFI has been shown to be an important regulator of cell migration in vivo during forebrain development (Alfano et al, 2011;Touzot et al, 2016;Tripodi et al, 2004;Zhou et al, 2015), but also in vitro and in cancerous cells (Adam et al, 2000;Boudot et al, 2014;Le Dily et al, 2008). Here, we show that it modulates the correct expression levels of the chemokine receptor gene Cxcr4 during GC neurogenesis and migration.…”
