Transcriptional Analysis of the EhPgp1 Promoter of Entamoeba histolytica Multidrug-resistant Mutant

Abstract: We present here the cloning and characterization of the EhPgp1 multidrug resistance gene promoter isolated from the Entamoeba histolytica drug-resistant mutant clone C2. The EhPgp1 promoter lacks the typical TATA box and the transcriptional initiation sequences described for other E. histolytica promoters. The major transcription initiation site of the EhPgp1 gene was located at the ATG start codon. The EhPgp1 core promoter located within the first 244 base pairs showed a higher chloramphenicol acetyltransfera… Show more

“…In E. histolytica, MDR phenotype is given mainly by overexpression of the EhPgp1 and EhPgp5 genes, which are finely regulated by transcriptional factors (7)(8)(9). Although EhPgp1 is constitutively expressed in drug-resistant trophozoites of clone C2, EhPgp5 gene is overexpressed only when C2 cells are grown in a high emetine concentration (10,11). Both genes are also amplified in the presence of a high drug concentration (12).…”

“…The 5Ј-flanking regions of a handful of E. histolytica genes have been studied via truncation and mutational analysis (8,(12)(13)(14). In the hgl2 promoter, a CCAAT-like motif was identified that decreased reporter gene expression when mutated, but the existence of proteins recognizing this element has not been demonstrated (3).…”

“…Just four promoters from the EhPgp1, EhPgp5, hgl5 and actin genes have been functionally characterized and until this moment have been functionally described the TATA box, the CAAT/enhancer binding elements, the GAACT and the UREs sequences [16][17][18][19][20][21][22][23][24]. With respect to the transcription factors have been reported the sequences for the TATA box binding protein (TBP) [25], two enhancer binding proteins (EhEBP1 and EhEBP2) that binds to the URE4 sequence [26] and the p53 [27] and C/EBP-like proteins (personal communication).…”
