Transcriptional Modulation of the Host Immunity Mediated by Cytokines and Transcriptional Factors in Plasmodium falciparum-Infected Patients of North-East India

Abstract: Complications due to malaria are caused mostly by host immunological responses. Plasmodium falciparum subverts host immunity by various strategies, including modulation in the host immune responses by regulating cytokines. The transcriptional alterations of major cytokines and immunoregulators were analyzed in this study through gene expression profiling in clinically defined subgroups of P. falciparum patients. Malaria patients were included from Dhalai district hospital of Tripura with uncomplicated malaria … Show more

“…These results propose that expression of these cytokines is not an immediate effect of parasitemia level and also suggest that the differences in cytokine concentrations among different groups may not be due to a direct effect of differences in parasite densities. Nevertheless, the fact that the association between cytokines and parasite densities was different among groups might be a reflection of diverse mechanisms of cytokine regulation depending on various other factors [ 63 ].…”

“…The experiments were completed by repeating twice. As an endogenous control, the Human Beta-actin gene was used with primers and cycling conditions, as described in [ 22 ]. Healthy control samples were used as calibrators, while a 3D7 strain of Pf and a confirmed field sample of Pv with genomic DNA concentration 5 ng/μL were used for standardization purposes.…”

“…). Although real-time PCR is considered gold standard for the detection of various pathogens because of their delightful sensitivity and specificity when tracing low target concentrations, 39 the natural characteristics of sgRNA in the identification of complementary target sequences provide it more specificity over primerbased detection systems. In addition, the simple and programmable nature of CRISPR/Cas technology can be combined with multiple signal detection platforms to enhance the technology applications through development of sensitive, specific, robust, and handy point-of-care diagnostic devices.…”
