Transcriptional Repression from the c-myc P2 Promoter by the Zinc Finger Protein ZF87/MAZ

Abstract: ZF87/MAZ is a zinc finger-containing transcription factor that was cloned based on its ability to bind to a site within the c-myc P2 promoter. However, its role in the control of c-myc transcription has not yet been well established. Here we have analyzed the effect of ZF87/ MAZ overexpression on transcription from the murine c-myc P2 promoter. It was found that when overexpressed in COS cells, ZF87/MAZ significantly represses transcription from P2. The repression is mediated through the ME1a2 element, located… Show more

“…Region V was the only other promoter region that showed an increase in the p300 and HDAC3 occupancy levels (3.3-and 9-fold, respectively). Regions IV and V contain binding sites for transcription factors MAZ (Me1a2; Izzo et al, 1999), E2F (Albert et al, 2001), CTCF (Filippova et al, 1996) that function as repressors and also TBP (TATA box; see Figure 2b). …”

“…Another cDNA with a high G/C content, the ZF87/ MAZ transcription factor (Izzo et al, 1999;Pyrc et al, 1992), was also used in this assay. Like the E2F1 cDNA, it also has an 80% G/C content within the ®rst 400 basepairs.…”

“…[18][19][20][21][22][23][24][25][26]55,56 The KRAB-domain represses the activity of CMV-promoter through direct communications with TATA box-dependent basal transcription machinery, 18,55 whereas mSIN3B interacts with essential DNAbinding proteins that place it in a promoter context in which it can repress transcription by recruiting proteins that condense chromatin by histone deacetylation. 19 In contrast to KRAB-A or mSIN3B, the Zik1-repressor domain does not repress gene transcription from the CMV gal -promoter and some of the other repressor domains showed only weak repression of CMV galactivity.…”
