Transcriptional Response of Silkworm (Bombyx mori) Eggs to O2 or HCl Treatment

Abstract: Diapause is a common biological phenomenon that occurs in many organisms, including fish, insects, and nematodes. In the silkworm (Bombyx mori), diapause generally occurs in the egg stage. Treatment with O2, HCl, or other compounds can prevent egg diapause. Here, we characterized the transcriptomic responses of newly laid eggs treated with O2 or HCl. Digital gene expression analysis showed that 610 genes in O2-treated eggs and 656 in HCl-treated eggs were differentially expressed. Of these, 343 genes were diff… Show more

“…Bivoltine silkworms (Dazao) were provided by the State Key Laboratory of Silkworm Genome Biology. As per the approach used in our previous study [ 13 ], diapause-destined eggs were obtained through the incubation of the parental generation under long-day conditions (18L/6D h) at 25 °C during embryonic development. Samples from diapause eggs were collected at 20 and 48 h oviposition, immediately frozen in liquid nitrogen, and stored at −80 °C until use.…”

“…Our previous studies on silkworm eggs via RNA sequencing showed that the gene expression patterns of hyperoxia- and HCl-treated diapause eggs were similar, which differed from non-treated diapause eggs [ 13 ]. Further physiological research has shown that reactive oxygen species (ROS) levels increase markedly in diapause eggs during this phase, but remain stable in non-diapause eggs, indicating that ROS may play a key role in promoting the onset of diapause [ 14 ].…”

“…6). It was reported that the rate of ribosome synthesis during diapause was lower than that of non-diapause eggs in Bombyx mori, so the upregulation of ribosomal proteins played an important role in blocking diapause [47,48] . In mosquitos, Culex pipiens, the expression of rpS3a was dramatically reduced for a short time in the diapause stage.…”
