Transcriptional Synergy between LIM-Homeodomain Proteins and Basic Helix-Loop-Helix Proteins: the LIM2 Domain Determines Specificity

Abstract: LIM-homeodomain proteins direct cellular differentiation by activating transcription of cell-type-specific genes, but this activation requires cooperation with other nuclear factors. The LIM-homeodomain protein Lmx1 cooperates with the basic helix-loop-helix (bHLH) protein E47/Pan-1 to activate the insulin promoter in transfected fibroblasts. In this study, we show that two proteins originally called Lmx1 are the closely related products of two distinct vertebrate genes, Lmx1.1 and Lmx1.2. We have used yeast g… Show more

“…We have previously described a similar effect of NLI in which NLI disrupted the transcriptional synergy between Lmx1.1 and E47 from the rat insulin I mini-enhancer by competing with E47 for direct interaction with the LIM domains of Lmx1.1 (16,40). In addition, it is possible that phosphorylation of the Isl1 C terminus blocks the interaction between Isl1 and Lhx3, further promoting the formation of NLI-dependent complexes.…”

“…Expression of Bacterial His 6 -tagged Recombinant Proteins-The cDNAs encoding hamster E47 (the hamster protein is usually called Pan1), mouse Beta2 (also known as NeuroD1), hamster PDX-1 (PDX-1 is also known as IPF-1, IDX-1, and STF-1), and hamster Lmx1.1 (46,47) were subcloned into pRSET-His 6 (Invitrogen). His 6 -tagged proteins were purified as described (48).…”

“…In support of this possibility, the Isl-1 knockout mouse was found to have no dorsal pancreatic mesenchyme or differentiated islet cells, despite normal expression of Pdx-1 in these animals (7). In addition, Isl-1, which is one of the LIM homeodomain proteins, may synergistically activate the insulin mRNA transcription rate through binding with basic helix-loophelix proteins (35,36). Together, these experiments and along with our results suggest that Isl-1 is a key factor for the differentiation of cells that produce insulin.…”
