Transcriptome analysis of fowl adenovirus serotype 4 infection in chickens

Abstract: Fowl adenovirus serotype 4 (FAdV-4) is a causative agent of inclusion body hepatitis and hydropericardium–hepatitis syndrome. These diseases cause considerable economic losses in the global poultry industry and are significant stressors for infected chickens. However, the molecular mechanisms of FAdV-4 pathogenesis are poorly understood. In the present study, we identified differentially expressed genes from the livers of FAdV-4-infected chickens using RNA-seq at 7, 14 and 21 days after FAdV-4 infection. We id… Show more

“…As a powerful research tool, transcriptome analysis has been used widely to reveal the interaction between host and virus. Based on this technology, several studies have revealed that changes in cell gene transcription are an important aspect of the interaction between FAdV-4 and its host [ 38 , 40 , 41 ]. It is worth noting that our previous study showed that the binding and entry of FAdV-4 into LMH cells profoundly affect early cellular miRNA transcription profiles and that cellular miRNAs play an important role in FAdV-4 entry [ 39 ].…”

“…In view of these findings, lncRNAs are an emerging hotspot in host–virus interaction research. Recently, several studies have investigated the roles of transcription products in the interaction between FAdV-4 and its host [ 38 , 39 , 40 , 41 ] by transcriptome analysis. However, the function of cellular lncRNAs during FAdV-4 infection is poorly understood.…”

“…Meanwhile, it is also involved in the immune response by mediating the NF-κB pathway and the NALP3 inflammatory pathway [33]. Ren et al [34] conducted transcriptome analysis on the liver of chickens infected with avian adenovirus type 4, and the results showed that P2RX7 was up-regulated after 21 days of infection. Similarly, Kalenik et al [35] showed that P2RX7 in spleen was significantly up-regulated after inoculation of chickens with three H5N1 subunit vaccines.…”

“…Compared to the mock-infected control, FAdV-4 infection depleted lymphocytes in the thymus and bursa through apoptosis and severe inflammatory response (Niu et al, 2019b). Analysis of the mRNA expression in livers of FAdV-4-infected chickens identified that IFN was downregulated at 7 days post-infection (Ren et al, 2019). However, the mechanisms of how FAdV-4 suppresses host innate immune response remain to be explored.…”
