Transcriptomic Analysis Reveals That Rho GTPases Regulate Trap Development and Lifestyle Transition of the Nematode-Trapping Fungus Arthrobotrys oligospora

Abstract: Nematode-trapping (NT) fungi are widely distributed in terrestrial and aquatic ecosystems. Their broad adaptability and flexible lifestyles make them ideal agents for controlling pathogenic nematodes. Arthrobotrys oligospora is a model species employed for understanding the interaction between fungi and nematodes.

“…The constructed vectors (pRS426-Ao PEX1 -hph and pRS426-Ao PEX6 -hph) were maintained in E. coli DH5α, and the target fragment for gene disruption was amplified using paired primers and transformed into A. oligospora protoplasts as described previously ( 47 , 53 ). Transformants were selected on PDAS (PDA supplemented with 0.6 mol/L sucrose) medium containing 200 μg/mL hygromycin ( 54 ). The putative transformants were confirmed by PCR amplification and Southern blot analyses (Fig.…”

“…For the staining of LDs, a glass slide was inserted into a PDA plate, and the fungal strains were incubated on PDA plates at 28 °C for seven days. Hyphal samples were stained with 10 µg/mL of boron dipyrromethene (BODIPY) dye (Sigma-Aldrich, St. Louis, MO, USA) for 30 min in the dark, and the LDs were observed by fluorescence microscopy [ 37 ]. The mycelia were examined using transmission electron microscopy (TEM; Hitachi, Tokyo, Japan).…”

“…Extracellular serine proteases are significant pathogenic factors in A. oligospora and other NT fungi during their infection of nematodes [ 26 , 27 , 28 ]. Since the genome of A. oligospora was sequenced in 2011, studies have focused on regulating mycelial growth, development, and pathogenicity of A. oligospora [ 29 , 30 , 31 , 32 , 33 , 34 , 35 ].…”
