Transcriptomics identified a critical role for Th2 cell-intrinsic miR-155 in mediating allergy and antihelminth immunity

Abstract: Significance The rising prevalence of allergic diseases throughout the world demands new approaches to treat this inflammatory disorder. CD4 + Th2 cells orchestrate the allergic cascade, stimulating IgE production, activating innate cells, and stimulating local tissue. This study took a comprehensive approach to identify the unique transcriptional features of pathogenic Th2 cells with the aim of identifying novel molecular regulators. Highly purified Th1, Th2, Th9… Show more

“…A separate report in human T-cell subsets also identified Blimp-1 and IFN-γ mRNAs as being targets of miR-125b, a signature miRNA of naïve CD4 + T cells responsible to block their differentiation into effector cells [9]. In sum, multiple miRNAs control IFN-γ and type 1 differentiation pathway (Table 1) Conflicting results were observed in an independent study by Wilson and colleagues, which used several in vivo models of Th2 response [67]. In this study miR-155 was essential to mount a protective Th2 response against house dust mites and helminths and lower levels of miR-155 in T cells led to a reduction in IL-13 production and reduced inflammation [67].…”

“…Dysregulated T H 2-cell responses following allergen exposure contribute directly to allergic disease (32). We recently reported that HDM-or helminthelicited T H 2 cells have distinct transcriptional profiles, compared with ex vivo T H 1, T H 17, and natural Treg cells (33). Using these transcriptional datasets (more than twofold change relative to naive T cells, P < 0.05) we applied in silico upstream regulator analyses (Ingenuity Pathways Analysis, IPA) (34) and generated a z-score representing the likelihood of activity of putative transcriptional regulators.…”

“…Cytokine and chemokine-secreting T H 2 cells orchestrate many of the pathophysiological features of allergic asthma. We therefore interrogated the transcriptional profile of highly purified T H 2 cells that contribute directly to HDM-induced airway allergy, in addition to T H 2 cells isolated from helminth-infected mice (33). Rather than focusing on differentially regulated genes in the datasets, we focused on putative upstream pathways that could contribute to the transcriptional profiles.…”
