Transforming Growth Factor β 1, Interleukin-8 and Interleukin-1, in Non–Small-Cell Lung Tumors

Abstract: A role in tumor progression has been proposed for transforming growth factor-beta 1 (TGF beta 1) and interleukin (IL)-8 as well as for IL-1, which itself induces the production of TGF beta 1 and IL-8 in many cell types. TGF beta 1 and IL-8 production and their regulation by IL-1 in five non-small-cell (NSC) lung tumor cell lines were evaluated. Moreover, their levels were evaluated in 29 NSC lung tumors. All cell lines constitutively produced TGF beta 1, and three produced IL-8. After IL-1 beta treatment, TGF … Show more

“…Overexpression of TGF-b1 in NSCLC compared to that of adjacent normal lung might play a role in BMP3B downregulation in NSCLC (Colasante et al, 1997). Similarly, BMP3, the closest family member of BMP3B, has also been shown to exert an antagonistic role against TGF-b1 in human bone marrow stoma during cell growth and differentiation (Faucheux et al, 1997).…”

“…In addition, we and others have noted that TGF-β1 can be produced by the tumor itself [17,[19][20][21][22][23]. Thus, since tumor size might dictate RT field size (and hence MLD), and since MLD may have an impact on TGF-β1, such analyses that consider each of these dosimetric, biologic, and tumor parameters are inherently complex.…”

“…Evidence of altered regulation of PTHrP mRNA in cancer cells was demonstrated in human lung cancer, which expressed all PTHrP mRNA isoforms in a pattern more similar to transformed keratinocytes (HaCaT) and HARA cells than the normal adjacent lung tissue, which expressed only the PTHrP 141 mRNA isoform. Lung cancers often express higher levels of TGF-1 mRNA (Colasante et al 1997), which may contribute to changes in PTHrP isoform expression between normal and cancerous tissue. Additional studies on the effect of TGF-1 on PTHrP mRNA isoform expression in lung tissue are ongoing in our laboratory.…”
