Transgene of MIF induces podocyte injury and progressive mesangial sclerosis in the mouse kidney

Abstract: Our results indicate that podocyte-expressed MIF could induce an injury of podocytes themselves, thereby accelerating the progression of glomerulosclerosis and leading to end-stage renal failure.

“…These cytokines mediate renal injury in experimental nephritis, including mesangial proliferative nephritis and anti-glomerular basement membrane antibody-induced glomerulonephritis [12,13]; however, the role of MIF in chronic renal diseases such as hypertensive and diabetic nephropathy has not been clarified.…”

“…The hypothesis is that the G/C substitution at 173 bp of the MIF gene increases the MIF level in serum and could therefore cause a pro-inflammatory response, induce injury to podocytes, and accelerate the progression of glomerulosclerosis [107]. TNF-α is also an important pro-inflammatory cytokine involved in the inflammatory process.…”

“…WT (B6, Japan SLC, Hamamatsu, Japan) and MIF Tg mice (Hokkaido University, Sapporo, Japan), (7) were used in this study. To detect the mif transgene, a pair of primers, CAGGS F1 5'-taatc aatta cgggg tcatt agttc atagc-3' and CAGGS B1 5'-tccca taagg tcatg tactg ggcat aatgc-3', were used for the amplification of a part of the vector sequence in the transgene (7). Polymerase chain reaction (PCR) amplification was performed with 1 cycle of 94˚C for 2 min, 35 cycles of 94˚C for 30 sec, 55˚C for 30 sec, 72˚C for 30 sec, and 1 cycle of 72˚C for 7 min.…”

“…After the molecular cloning of the mif gene (2), it has become increasingly clear that MIF is a unique cytokine with various actions on many target cells (3,4). Recent studies, using genetically engineered animals such as MIF knockout (5,6) and MIF transgenic (MIF Tg ) (7,8) mice, have revealed the importance of MIF in inflammatory diseases.…”
