Transplantation of Embryonic Neural Stem/Precursor Cells Overexpressing BM88/Cend1 Enhances the Generation of Neuronal Cells in the Injured Mouse Cortex

Abstract: The intrinsic inability of the central nervous system to efficiently repair traumatic injuries renders transplantation of neural stem/precursor cells (NPCs) a promising approach towards repair of brain lesions. In this study, NPCs derived from embryonic day 14.5 mouse cortex were genetically modified via transduction with a lentiviral vector to overexpress the neuronal lineage-specific regulator BM88/Cend1 that coordinates cell cycle exit and differentiation of neuronal precursors. BM88/Cend1-overexpressing NP… Show more

“…CYP2W1 [29], GPR146 [30] and TOP1MT [31] are important for nutrient metabolism in the intestine. CEND1 plays a role in neuronal differentiation [32], thus in development of the enteric nervous system. The down-regulation of these genes due to promoter hypermethylation may significantly affect intestinal metabolism and development, which may help to explain the difficulties of preterm neonates to tolerate enteral feeding and the down-regulation of proteins involved in intestinal metabolism and motility [8].…”

“…It is well known that increased neuronal differentiation is helpful to the functional recovery after CNS insults [3]. Besides, another potentially beneficial effect of miR-17-92 cluster-overexpressing NSCs is the significant inhibition of astrogliosis since reactive astrocyte as well as glial scar may have detrimental consequences after CNS damage [48, 49].…”

“…They play essential roles in repairing and maintaining normal CNS cells. Besides, tissue engineering by normal NSCs transplantation also enables the clinical therapeutic applications for CNS regressive diseases, such as Alzheimer's, Parkinson's, Huntington's disease and other CNS disorders (Storch and Schwarz, 2002;Lee et al, 2005;Oliveira and Hodges, 2005;Sanberg, 2007;Tang et al, 2008;Moghadam et al, 2009;Zhongling et al, 2009;Makri et al, 2010).…”
