TRAT1 overexpression delays cancer progression and is associated with immune infiltration in lung adenocarcinoma

Abstract: The roles and mechanisms of T-cell receptor (TCR)-associated transmembrane adaptor 1 (TRAT1) in lung adenocarcinoma (LAC) have not yet been reported in the relevant literature. Therefore, this study aimed to understand the roles and mechanisms of TRAT1 in LAC using bioinformatics and in vitro experiments. TRAT1 expression levels in LAC samples were analysed using various databases. TRAT1 co-expressed genes were acquired by the correlation analysis of LAC tissues. The functional mechanisms and protein network o… Show more

“…In humans, defects in the CD3D, CD3E and CD3Z genes are a cause of severe immunode ciency and appear early in life and increase susceptibility to infection [16]. Overexpression of T cell receptor (TCR)-related transmembrane junction 1 (TRA1) inhibits the growth and migration of lung adenocarcinoma cells, thereby delaying cancer progression [17]. We have concluded from bioinformatic analyses that CDs are not a major source of cancer.…”

“…IL-4 is capable of triggering cytotoxic responses in IL-17-producing CD8+ T cells (Tc17 cells), with IL-4/AKT signaling inducing TRAT1 upregulation and TCR stabilization in these Tc17 cells, thus enhancing their cytotoxic activity ( 29 ). TRAT1 expression is closely related to immune activity and the degree of infiltration by a range of immune cell populations including CD8+, cytotoxic, Th1, Th17, and Treg cells ( 30 ).…”

“…In 6 well plate, the clean tip of pipette is used to create “wound” when cells form confluent monolayer, and cells cultured in RPMI-1640 were supplemented with 0% fetal bovine serum. Transfected cells were used to capture the image of wound edge with optical microscope at 0 h. After 24 h of incubation, the cell images of the same area were captured for measurement, and the wound healing rate was calculated [ 19 ]. Transfected cells were investigated for transwell migration.…”

“…According to the manufacturer's agreement of transwell System, fix the transwell chamber with 5% paraformaldehyde for 15 min, and then dye it with 0.1% crystal violet for 10 min. Transmembrane cells were calculated under inverted fluorescence microscope [ 19 ].…”
