Triamcinolone acetonide protects auditory hair cells from 4-hydroxy-2,3-nonenal (HNE) ototoxicity in vitro

Abstract: Triamcinolone acetonide did not affect hair cell integrity in the organ of Corti explants and it provided a high level of protection of hair cells against the ototoxic effects of a damaging level of HNE as determined by hair cell density counts.

“…The protective effect of TA against oxidative stress in hair cells has been reported [38]. In ophthalmic field, TA is known to reduce VEGF expression from RPE cells [39], and thus the permeability of retinal vascular endothelium (inner BRB) is reduced, which may be followed by the reduction of edema.…”

“…Control cultures consisted of cochleae that were left untreated or were treated with lipofected scrambled siRNA (scRNALF) or scRNA-containing NPs (scRNANP). 4-HNE was prepared as a 60 mM stock solution in DMSO and used at a final concentration of 100-475 μM (Guzman et al, 2006; Ruiz et al, 2006). …”

“…To estimate HC numbers, we counted hair bundles and/or cuticular plates under 40X magnification (Olympus BX51, Melville, NY). In the cochleae, two regions from each turn (basal and middle turns, 100 μm in each region ≈ 1.67% of basilar membrane length) were randomly chosen, and the number of inner and outer HCs (IHCs and OHCs, respectively) within each region was estimated and statistically analyzed (Guzman et al, 2006; Ruiz et al, 2006; Takebayashi et al, 2007). In the maculae, four to five areas (50×50 μm 2 per area) were randomly chosen from the striolar and edge regions, and the number of HCs within each area was estimated and statistically analyzed.…”

“…4-hydroxy-2,3-nonenal (HNE) (Camandola et al, 2000). HNE has also been shown to be formed in the cochlea in response to excessive noise exposure (Yamashita et al, 2004) and to directly kill auditory hair cells when applied to 4-day old (P-4) rat organ of Corti explants (Guzman et al, 2006). The reduced form of glutathione (GSH) can bind to HNE and inactivate it (Bosch-Morell et al, 1999).…”
