Trout P450IA1: cDNA and Deduced Protein Sequence, Expression in Liver, and Evolutionary Significance

Abstract: The cytochrome P1(450) (P450IA1) cDNA has been isolated and sequenced from liver of 3-methylcholanthrene-treated rainbow trout (Salmo gairdneri). The cDNA hybridizes to a 2.8-kb mRNA that is induced at least 10-fold by 3-methylcholanthrene. Southern blot analysis suggests the presence of a single gene or a very small number of genes. An open reading frame of the 2573-bp cDNA encodes a 522-residue protein (Mr = 59,241) that is more similar to the mammalian P450IA1 than the mammalian P450IA2 proteins. The aromat… Show more

“…Lesions (9,124), and (e) many liver and skin lesions seen in epizootics are similar to those classically described in rodent carcinogenesis. In addition, the resistance of related species in similar habitats to the development of neoplasms indicates differing species susceptibilities (33,134 (84,212), scup (Stenotomus chrysops) (104,105), cod (Gadus morhua) (65), English sole (158), and other species (185). The major Phase II routes of metabolism for electrophilic and nucleophilic compounds in fishes are glutathione and glucuronic acid conjugation, respectively (64).…”

“…CYP1A nucleotide sequences have been determined in rainbow trout (Berndtson and Chen, 1994;Heilmann et al, 1988), plaice (Leaver et al, 1993), Atlantic tomcod (Roy et al, 1995), toadfish and scup (Morrison et al, 1995), killifish (Morrison et al, 1998), red sea bream (Mizukami et al, 1994), sea bass (Stien et al, 1998) and, more recently, Atlantic salmon (Rees et al, 2003). Current evidence shows that several CYP1A isoforms exist in fish (Teramitsu et al, 2000) and they are distributed in tissues such as the liver, gut, kidney, gill and heart (Goksøyr and Husoy, 1998;Sarasquete and Segner, 2000;Stegeman and Hahn, 1994).…”
