Tubeimoside-1 suppresses breast cancer metastasis through downregulation of CXCR4 chemokine receptor expression

Abstract: To examine the effect of TBMS1on breast cancer metastasis, and investigate the potential mechanism by which Tubeimoside-1 (TBMS1) inhibits the CXCR4 expression in breast cancer cells. The expression of CXCR4 in breast cancer cell lines was determined by immunoblotting and real-time PCR. The effect of TBMS1 on NF-κB binding activity was evaluated by EMSA assay and ChIP analysis. Cell proliferation and invasion were analyzed by MTT assay and transwell invasion assay, respectively. The effect of TBMS1 on breast c… Show more

“…Viability of cells for the extracts (1, 2.5, 5, 10, 25, 50, 100, and 200 lg/mL), compounds 1 -8 (1, 2.5, 5, 10, 25, 50, 100, and 200 lM), and L-NMMA (50, 100, 200, 400, and 800 lM) as a positive control was determined by an Ez-Cytox cell viability detection kit purchased from Dail Lab Service Co. (Seoul, Korea), as reported previously. [20] [21] Measurement of Nitric Oxide Production RAW264.7 cells were plated into a 96-well plate at a density of 3 9 10 4 cells/well and preincubated at 37°C for 24 h. After serum starvation for 12 h, cells were pretreated with the extracts (1, 2.5, 5, 10, 25, 50, 100, and 200 lg/mL), compounds 1 -8 (1, 2.5, 5, 10, 25, 50, 100, and 200 lM), and L-NMMA (50, 100, 200, 400, and 800 lM) as a positive control for 1 h, and then stimulated with 100 ng/mL of LPS for another 24 h. To evaluate NO levels in the culture media, nitrite, a soluble oxidation product of NO, was measured using the Griess reaction. [30] [31]…”

“…In vitro invasion/migration assays were performed using a 24-well Transwell unit with polycarbonate filters (Corning Life Sciences) as previously described (26). The lower side of the filter was coated with type I collagen (Sigma-Aldrich) and the upper side of the filter was coated with Matrigel (BD Biosciences, Bedford, MA, USA).…”

“…Tubeimoside-1 has capability to effectively inhibit apoptotic signaling molecule NF-κB via its upstream signaling factor TNF-α modulation in HepG2, A549, and PC9 cells [36,46]. Tubeimoside-1 down-modulated the expression of CXCR4 (a chemokine receptor which promotes tumor metastasis) in breast cancer cells by suppressing NF-κB binding activity to CXCR4 promoter as various transcriptional factors such as NF-κB, PPAR-γ, and HIF-1α mediate the up-regulation of CXCR4 in cancerous cells [67]. After exposure [68,69].…”
