Tumor-associated macrophages and anti-tumor therapies: complex links

Abstract: Myeloid cells infiltrating the tumor microenvironment, especially tumor-associated macrophages (TAMs), are essential providers of cancer-related inflammation, a condition known to accelerate tumor progression and limit the response to anti-tumor therapies. As a matter of fact, TAMs may have a dual role while interfering with cancer treatments, as they can either promote or impair their functionality. Here we review the connection between macrophages and anticancer therapies; moreover, we provide an overview of… Show more

“…Particularly, they promote tumor growth and dissemination, enhance angiogenesis, contribute to matrix degradation, and suppress anti‐tumor Th1‐mediated adaptive immunity (Chittezhath et al, ; Squadrito & De Palma, ). TAMs are major source of reactive mediators such as cytokines, chemokines, growth factors, ROS and RNS, and proteolytic enzymes (Belgiovine et al, ; Chittezhath et al, ). Pharmacological depletion of TAMs results in an inhibition of angiogenesis in tumors (Squadrito & De Palma, ).…”

“…It is induced by myeloid regulatory cells such as neutrophils [212,213], TAM [214,215,216], MDSC [217,218,219,220], and regulatory DC [221,222]. In addition, Treg and carcinoma-associated fibroblasts (CAF) contribute to the immunosuppressive milieu [222,223,224,225].…”

“…According to the literature, tumor associated macrophages (TAMs) are the major infiltrating leukocytes of the tumor micro-environment [62]. They have usually a M2-like phenotype and an immunosuppressive activity, producing growth factors for the tumor cells [62].…”

“…According to the literature, tumor associated macrophages (TAMs) are the major infiltrating leukocytes of the tumor micro-environment [62]. They have usually a M2-like phenotype and an immunosuppressive activity, producing growth factors for the tumor cells [62]. TAMs also promote dissemination of tumor cells enhancing angiogenesis and matrix degradation, suppressing Th1 immne ativity [63].…”
