Tumor-Associated Microglia and Macrophages in the Glioblastoma Microenvironment and Their Implications for Therapy

Abstract: Glioblastoma is the most frequent and malignant primary brain tumor. Standard of care includes surgery followed by radiation and temozolomide chemotherapy. Despite treatment, patients have a poor prognosis with a median survival of less than 15 months. The poor prognosis is associated with an increased abundance of tumor-associated microglia and macrophages (TAMs), which are known to play a role in creating a pro-tumorigenic environment and aiding tumor progression. Most treatment strategies are directed again… Show more

“…Indeed, GBM cells recruit microglia and macrophages in the TME by means of chemoattractants (e.g., C-C motif chemokine ligand 2 (CCL2), CSF-1) [ 69 , 70 ] and induce a switch to pro-tumoral subtype [ 65 , 71 , 72 ]. In return, TAMs promote tumor cell proliferation and angiogenesis as well as inhibition of effector T cells proliferation and attraction of T regulatory cells (Tregs) and MDSCs through cytokines secretion [ 65 , 72 , 73 ].…”

“…Particularly, tumor-associated macrophages (TAMs) are the dominant infiltrating immune cell population [32]. TAMs display a heterogeneous spectrum of phenotypes, of which extremes exert either antitumor (M1-like) or protumor functions (M2-like) [33].…”
